a summary Lithium is a first-line treatment for bipolar affective disorder, and it has recently been shown to have anti-disease Alzheimer’s effects in populations at risk (AD). We discovered that NP03 prevented the onset of AD pathology by inactivating GSK-3, lowering BACE1 expression and activity, and lowering amyloid levels. Specifically, NP03 improved memory loss, synaptic plasticity gene CRTC1 promoter binding, and hippocampal neurogenesis. These findings suggest that NP03 could have therapeutic potential in the early or preclinical stages of Alzheimer’s disease.

This paper examines systematic clinical findings that indicate memory and cognitive dysfunction in patients treated with lithium for unipolar and bipolar affective disorders. However, a number of studies have failed to show that lithium causes memory problems. As a result, the findings of studies were ambiguous. The heterogeneity of samples, as well as a number of methodological and design issues, contributed to the lack of scientific consensus. Short- and long-term memory definitions were often subjective and lacked common requirements. Some research found that memory test scores remained stable over time and that subjective complaints of memory failure were linked to depression. Lithium’s cognitive side effects, on the other hand, are a major concern and a major source of patient pain, leading to partial and/or complete non-adherence to medication. Lithium has been linked to memory loss, difficulty finding words, and recall problems. My patients have often reported cognitive “dulling” and a lack of cognitive “creativity” as side effects of lithium treatment. This seems to be dose-dependent.
