The qiagen revolution

The RNeasy Mini Kit uses silica-membrane RNeasy spin columns with a binding capacity of 100 g RNA to quickly purify high-quality RNA from cells, tissues, and yeast. Tissue samples can be easily stabilized with RNAprotect Tissue Reagent or Allprotect Tissue Reagent, and disturbed with a TissueRuptor or TissueLyser. The QIAcube Connect will completely automate RNA purification using the RNeasy Mini Kit. The RNeasy Micro Kit (spin-column binding capacity of up to 45 g RNA), RNeasy Midi Kit (spin-column binding capacity of 1 mg RNA), and RNeasy Maxi Kit (spin-column binding capacity of 6 mg RNA) are also available for smaller and larger samples.
RT-PCR of total RNA isolated from the indicated number of HeLa cells using the RNeasy Mini Pack. 10 l (1/5) of the eluate was digested with RNase-free DNase before being reverse transcribed with an oligo-dT primer. In a 50 l PCR, 2.5 l (1/20) of the cDNA mix was used. A 452-bp GAPDH fragment was amplified. C- denotes a negative power, C+ denotes a positive control, and M denotes a 100-bp ladder.

First steps of dna extraction with a commercial kit

The QIAamp DNA Blood Mini Kit purifies DNA using a silica membrane. The QIAamp DNA Blood Mini Kit can filter up to 200 liters of fresh or frozen whole blood. In a centrifuge or on vacuum manifolds, QIAamp Mini spin columns can be easily processed. The QIAcube Connect will completely automate DNA purification using the QIAamp DNA Blood Mini Kit.
Depending on the age and storage of the samples, QIAamp DNA Blood Kits yield DNA ranging from 200 bp to 50 kb (see figure “Apoptotic banding in stored blood”). Purified DNA can be used for long-range PCR amplification (see “Long-range PCR”) and restriction fragment length polymorphism (RFLP) analysis, which can be used for paternity testing, for example (see figure “Paternity testing by RFLP analysis”).
There is no need for phenol–chloroform extraction. While pollutants move through, DNA binds directly to the QIAamp silica-gel membrane. In two efficient wash steps, PCR inhibitors such as divalent cations and proteins are completely extracted, leaving only pure nucleic acid to be eluted in either water or a buffer supplied with the package. The QIAamp DNA Blood technology extracts genomic, mitochondrial, or viral DNA from blood and other body fluids, ready for PCR and blotting.

The QIAamp DNA Microbiome Kit is a dedicated solution for bacterial microbiome DNA purification and enrichment from swabs and body fluids. During the purification process, successful depletion of host DNA maximizes bacterial DNA coverage in next-generation sequencing analysis, allowing for highly sensitive 16S rDNA-based microbiome analysis and whole metagenome shotgun sequencing studies.
The V4 region was amplified using [A] DNA (10 ng) purified from buccal mucosa swabs using the QIAamp DNA Microbiome Kit or kits from three other suppliers. Amplification of samples isolated with the QIAamp DNA Microbiome protocol was most effective after 25 cycles, demonstrating the advantage of removing host DNA. [B] The V4 amplicons of two samples purified with the QIAamp DNA Microbiome Kit were mapped to reference genomes derived from the Ribosomal Database Project after sequencing on the Illumina MiSeq platform. The bacterial makeup was similar to that of the human oral microbiome.
The QIAamp DNA Microbiome Kit’s tailored protocol eliminates bias caused by microbe lysis susceptibility differences. Six cultured bacterial species were applied to buccal swab samples to create a model microbiome. Plate counts were used to determine the composition of the population. The Illumina MiSeq platform was used to conduct whole metagenome shotgun sequencing, and the output reads were compared to the input numbers. The QIAamp DNA Microbiome Kit was used to prepare the samples, which allowed for the most accurate relative composition.
